通过对白屈菜低温应答过程的转录组分析发现膜脂不饱和化相关基因的表达在一定过程中发生变化,脂肪酸去饱和酶基因FAD2在随温度的变化趋势为正"V"型,且表达量变化显著。利用NCBI等在线软件对序列进行相关生物学信息分析,并对白屈菜FAD家族成员FAD2基因的完整开放阅读框(ORF)进行克隆,并命名为CmFAD2。选用克隆载体pMD-19-T,转化大肠杆菌DH5α,测序验证序列正确性及完整性。将目的基因与植物表达载体pRI-201-AN连接构建重组DNA pRI-201-AN-Cm FAD2,电击法转化农杆菌LBA4404,利用菌液PCR法验证成功。该基因可作为药用植物抗寒品种创制的候选基因。 相似文献

AIM: To observe the effect of beclin-1 silencing by the technique of RNA interference on the injury of human gastric cancer SGC-7901 cell by Sheliugu extract (the extract from tuber of Amorphophallus konjac, TuAKe). METHODS: To knock down the expression of beclin-1 gene, SGC-7901 cells were transfected with lentiviral vector carrying beclin-1-shRNA. The beclin-1 gene knock-down and non-knock-down SGC-7901 cells were treated with TuAKe. The cell viability was analyzed by CKK-8 assay. The percentages of apoptotic cells were detected by flow cytometry. The expression of beclin-1 and LC3 was detected by Western blot. RESULTS: The beclin-1 gene silencing decreased the protein expression of beclin-1 and increased the protein expression of LC3 in the SGC-7901 cells, leading to the decrease in cell viability and the increase in apoptotic rate (P<0.05). TuAKe increased the protein expression of beclin-1 and LC3 in the SGC-7901 cells, and decreased the protein expression of LC3 in the SGC-7901 cells with beclin-1 gene silencing, thus inhibiting the cell viability and increasing the apoptotic rate (P<0.05). CONCLUSION: Beclin-1 gene silencing inhibits the activation of beclin-1-related signaling pathway in gastric cancer SGC-7901 cells, and aggravates the injury of cell viability induced by TuAKe. 相似文献

AIM:To investigate the effect of proline-spirooxindole on the viability and apoptosis of human non-small-cell lung cancer A549 cells. METHODS:The effect of proline-spirooxindole on the viability of A549 cells was determined by CCK-8 assay. The apoptosis was analyzed by flow cytometry. The effects of proline-spirooxindole on the expression of PARP and p53 and the phosphorylation of mTOR were determined by Western blot. RESULTS:After A549 cells were treated with proline-spirooxindole (25, 50 and 100 mg/L), the cell viability was decreased (P<0.01) compared with DMSO control group. The apoptotic rate was increased compared with DMSO control group (P<0.01). The protein expression of p53 was up-regulated, the increased apoptotic protein cleaved PARP was observed, and the phosphorylation of mTOR was inhibited (P<0.01). CONCLUSION:Proline-spirooxindole inhibits the viability of A549 cells and induces apoptosis, which may be related to the phosphorylation of mTOR. 相似文献

基于部首嵌入和注意力机制的病虫害命名实体识别 总被引:1,自引:0,他引:1
为了解决农业病虫害命名实体识别过程中存在的内在语义信息缺失、局部上下文特征易被忽略和捕获长距离依赖能力不足等问题,以农业病虫害文本为研究对象,提出一种基于部首嵌入和注意力机制的农业病虫害命名实体识别模型(Chinese agricultural diseases and pests named entity recognition with joint radical embedding and self attention, RS-ADP)。首先,该模型将部首嵌入集成到字符嵌入中作为输入,用以丰富语义信息。其中,针对部首嵌入设计了3种特征提取策略,即卷积神经网络(Convolutional neural network, CNN)、双向长短时记忆网络(Bidirectional long short term memory network, BiLSTM) 和CNN-BiLSTM;其次,采用多层不同窗口尺寸的CNNs层提取不同尺度的局部上下文信息;然后,在BiLSTM提取全局序列特征的基础上,采用自注意力机制进一步增强模型提取更长距离依赖的能力;最后,采用条件随机场(Conditional random field, CRF)联合识别实体边界和划分实体类别。在包含11个类别和24715条标注样本的农业病虫害自制语料上进行了实验。结果表明,本文模型RS-ADP在该数据集上精确率、召回率和F1值分别为94.16%、94.47%和94.32%;在具体实体类别上,RS-ADP在作物、病害、虫害等易识别实体上F1值高达95.81%、97.76%和97.23%。同时,RS-ADP在草害、病原等难以识别实体上F1值仍保持86%以上。实验结果表明,本文所提模型能够有效识别农业病虫害命名实体,其识别精度优于其他模型,且具有一定的泛化性。 相似文献

The objective of this study was to investigate the characteristics of ruminal microbial communities of alpacas (Lama pacos) and sheep (Ovis aries) fed three diets with varying ratios of roughage (corn stalk) to concentrate, 3:7 (LS), 5:5 (MS) and 7:3 (HS). Six alpacas (one-year-old and weighing 29.5 ± 7.1 kg) and six sheep (one-year-old and weighing 27.9 ± 2.7 kg) were used in this study, in a replicated 3 × 3 Latin square experiment. Total protozoa concentration was determined under the microscope; total fungi and methanogens were assessed using quantitative polymerase chain reaction and expressed as a percentage of total bacterial 16S rRNA gene copies; bacterial communities were investigated by targeted 16S rRNA gene (V3–V4 region) sequencing. The percentage of fungi was significantly higher in alpacas than in sheep under the LS diet, while the concentration of protozoa was significantly lower in alpacas under HS, MS and LS diets. The alpha diversity including Shannon, Chao l and ACE indices of bacterial communities was higher in alpacas than in sheep, under the LS diet. A total of 299 genera belonging to 22 phyla were observed in the forestomach of alpaca and sheep, with Bacteroidetes and Firmicutes dominating both animal species. Phyla Armatimonadetes and Fusobacteria, as well as 64 genera, were detected only in alpacas, whereas phyla Acidobacteria and Nitrospira, as well as 44 genera, were found only in sheep. The abundance of cellulolytic bacteria, including Butyrivibrio and Pseudobutyrivibrio, was higher in alpacas than in sheep under all three diets. These differences in the forestomach microbial communities partly explained why alpacas displayed a higher poor-quality roughage digestibility, and a lower methane production. Results also revealed that the adverse effects of high-concentrate diets (70%) were lesser in alpacas than in sheep. 相似文献
